Which of the File Managers is best for accessing/managing files to & from an external storage device. I have an OTG cable and USB memory stick which I would like to manage using a suitable File Manager. Will either ES Explorer of Astro do the trick.

Without "Root", you will not be able to write to external storage, regardless of your choice of file manager. To write to to external storage you will need Root and the app, "Stickmount." Notably, there is a paid app that will allow you to read from external storage by OTG, without Root and a free app that allows limited viewing.

The developer gave me a beta version of Nexus Media Importer to test. It allows writing to FAT without root. Not sure when the version 5 will be available at the store but should be soon.

Cool! It is also possible to write and read the usb stick by plugging it into a computer on the same wifi network, enabling share for that drive or a backup folder on the hard drive, and using es file explorer for example to transfer the files over wifi.

Sure. Try one of the wifi file transfer apps, other than the one by smarterdroid. They crippled that one to a 4Mb file size.
I'm not the lowest of the low, but I am the slowest of the slow (49.5 mpg over 42k miles in Scion xB)

I have Nexus Media Importer which works well but doesn't seem to be able to export (I suppose the name is a giveaway).
Don't want to root my Nexus as I'm afraid that I might do something to kill it.
